There will be buses certainly but you might have to get to Madrid first on the way to Avila and Toledo. No problem since the bus system is good enough, unless connection time is a key factor for you. If connection time is not an issue then you are ok.
I don't know the best way from Granada to Barna but both are amongst the main touristic places in Spain and you should have no problem for that either. Roads and coaches are good enough in Spain and smoking in coaches is forbidden. Fares are not expensive.
